SUMMARY: Recently, several methods for analyzing phenotype data have been published, but only few are able to cope with data sets generated in different studies, with different methods, or for different species. We developed an online-system in which more than 300,000 phenotypes from a wide variety of sources and screening methods can be analyzed together. Clusters of similar phenotypes are visualized as networks of highly similar phenotypes, inducing gene groups useful for functional analysis. This system is part of PhenomicDB, providing the world's largest cross-species phenotype data collection with a tool to mine its wealth of information. Hepatobiliary elimination via canalicular efflux transport proteins plays a key role in the clearance of endo- and xenobiotics. Correct membrane localization and coordinated action of the transport systems are essential for vectorial transport of drugs from blood into the bile. Whilst basolaterally localized uptake transporters are responsible for the inward transport of substances from the blood into the hepatocyte, apically expressed ATP-dependent transport proteins such as Pglycoprotein (P-gp), multidrug resistance-associated protein (Mrp2) and breast cancer resistance protein (Bcrp) mediate the outward efflux into the bile canaliculus. Using sandwich-cultured rat hepatocytes we have (…)text/htmlhttp://www.ncbi.nlm.nih.govNCBI PubMed18.06.2010 | NCBI PubMed: Cholesteryl ester transfer protein and its inhibition.

Cholesteryl ester transfer protein (CETP) is a plasma glycoprotein that facilitates the transfer of cholesteryl esters from the atheroprotective high density lipoprotein (HDL) to the proatherogenic low density lipoprotein cholesterol (LDL) and very low density lipoprotein cholesterol (VLDL) leading to lower levels of HDL but raising the levels of proatherogenic LDL and VLDL. Inhibition of CETP is considered a potential approach to treat dyslipidemia. However, discussions regarding the role of CETP-mediated lipid transfer in the development of atherosclerosis and CETP inhibition as a potential strategy for prevention of atherosclerosis have been controversial. Scientists from the German Cancer Research Center have investigated which genes are responsible for this survival strategy of cancer cells. The revealed that cancer cells rely on the tension of specific protein fibers to be able to multiply. Thus, proteins which maintain this tension are promising targets for new, target-specific anticancer drugs: If they are switched off, cancer cells die. Patients with bone metastases from renal cell carcinoma suffer from severe pain, nerve compression syndromes and pathologic fractures. Very little is known about the mechanisms of skeletal metastases of RCC. Thus, to better understand the molecular mechanism of renal cell cancer (RCC) bone metastasis, it is crucial to develop new animal models. We have established a new animal model of RCC metastasis to bone by inoculation of human 786-O/luciferase cells into the left cardiac ventricle (…)text/htmlhttp://www.businesswire.comBusiness Wire04.05.2010 | Business Wire: Research and Markets: German Pharma Sector Analysis | Business Wire

Sorafenib (Nexavar) is a novel oral Raf kinase and vascular endothelial growth factor receptor inhibitor. Most anti-cancer drugs are substrates for ABC efflux pumps especially for P-glycoprotein (P-gp). To evaluate the influence of P-gp on the pharmacokinetics of sorafenib substrate properties for this transporter were investigated. Therefore, permeability of sorafenib across Caco-2 and P-gp overexpressing cells was determined. To determine the in vivo relevance of these in vitro findings, pharmacokinetics of sorafenib in mdr1a/1b(-/-) and wild-type (WT) mice was studied. Sorafenib is highly permeable and exhibits a slight efflux across Caco-2 cells. In P-gp overexpressing cells a small (…)text/htmlhttp://www.businesswire.comBusiness Wire26.04.2010 | Business Wire: Research and Markets: China Pharmaceuticals and Healthcare Report Q2 2010 | Business Wire
